How do mammalian mitochondria synthesize proteins?

چکیده
Mitochondria contain their own genome that is expressed by nuclear-encoded factors imported into the organelle. This review provides a summary of the current state of knowledge regarding the mechanism of protein translation in human mitochondria and the factors involved in this process.

Differential regulation of full-length genome and a single-stranded 7S DNA along the cell cycle in human mitochondria
Mammalian mitochondria contain full-length genome and a single-stranded 7S DNA. Although the copy number of mitochondrial DNA (mtDNA) varies depending on the cell type and also in response to diverse environmental stresses, our understanding of how mtDNA and 7S DNA are maintained and regulated is limited, partly due to lack of reliable in vitro assay systems that reflect the in vivo functionali...
متن کاملOvercoming stalled translation in human mitochondria
Protein synthesis is central to life and maintaining a highly accurate and efficient mechanism is essential. What happens when a translating ribosome stalls on a messenger RNA? Many highly intricate processes have been documented in the cytosol of numerous species, but how does organellar protein synthesis resolve this stalling issue? Mammalian mitochondria synthesize just thirteen highly hydro...
متن کاملSynthesis of cytochrome oxidase components in isolated mitochondria of Neurosora crassa.
The formation of polypeptides within the mitochondrion has been documented by a large body of evidence. The ability of isolated mitochondria to incorporate amino acids into membrane protein is an expression of this cellular capacity (for reviews see [ l-61 ). However, until now it has not been demonstrated that isolated mitochondria are able to synthesize definite polypeptides which are compone...
متن کاملLactate Metabolism is Associated with Mammalian Mitochondria
It is well established that lactate secreted by fermenting cells can be oxidized or used as a gluconeogenic substrate by other cells and tissues. It is generally assumed, however, that within the fermenting cell itself, lactate is produced to replenish NAD+ and then is secreted. Here we explore the possibility that cytosolic lactate is metabolized by the mitochondria of fermenting mammalian cel...
متن کاملRegulation of Mitochondrial Membrane Assembly in Neurospora crassa
Cultures of mutant cni-1, a chromosomal mutant of Neurospora crassa, undergo a marked change in respiratory properties as the age of the culture increases. Early log phase cultures have a high level of respiration that is insensitive to inhibition by cyanide or antimycin A. Late log and stationary phase cultures have reduced rates of respiration. A high percentage of this respiration is inhibit...
